Abstract

ABSTRACTGenetic Variation of Monascus purpureus Isolated from Chinese Red Rice. An attempt was carried out to detect genetic variation among six Monascus purpureus isolates by RAPD finger printing. The isolates studied were isolated from Chinese red rice (ang-kak) collected from Indonesian markets. Seven types of oligonucleotide 12-mer primers were used in this study. These primers were different from G+C ratio (42%, 50%, 68% and 75%) so as to know the best ratio for DNA amplication. Products of DNA amplication by seven primers were observed such as 158 banding patterns, comprise of 23 common fragments and 135 polimorfic fragments. The result showed that the six isolates were divided into two groups based on DNA banding patterns. The same grouping was also shown by phylogenetic analysis. This analysis indicated two cluster which were different from its genetic distance (49%) and two pairs of isolates were identical to each other. So far, this RAPD analysis proved that there was high genetic variation within the six isolat of M. purpureus. It was assumed that there was a tendency of species separation within those isolates. It is suggested that sequencing analysis on 18S rDNA should be carried out to ensure status of M.purpureus species used in ang-kak production.Key words: Monascus purpureus, variation, genetic, Chinese red-rice, RAPD

Abstract

ABSTRACTCharacterization of Amylase Production by Saccharomyces cerevisiae W303A Recombinants. Cloning of amylase gene from Endomycopsis fibuligera ITB.R.cc.64 into S. cerevisiae W303a can effectively increase the yeast function to digest starch directly into ethanol. Production of amylase by S. cerevisiae W303a recombinants (I and P) were done by growing in yeast peptone starch (YPS) medium. The result showed that the recombinants could be produced of amylase by gave clear zone after staining by iodium vapor. The optimum condition of production of amylase by S. cerevisiae W303a recombinants were pH 7.0, 40?C temperature incubation, and gave maximum activity after 36 hours incubation. Amylase activity of I was higher than P recombinant for these condition respectively.Key words: Characterization, amylase, S. cerevisiae W303a

Abstract

ABSTRACTBiological Activity of Enzymatically Synthesized Polyphenol Glycoside on Microbial Growth. We have studied an indigenous bacterial strain produced a glycosyl transfer enzyme (CGTase) yielding polyphenol glycosides from a substrate of starch and polyphenol-aglycone. We observed that the CGTase derived from culture filtrate of some microbial strains (Candida rugosa, Bacillus megaterium, B. coagulans and B. polymixa) could synthesize transfer products in the presence of appropriate polyphenol-aglycones as their acceptors. An inhibitory effects of enzymatically synthesized polyphenol glycosides against bacterial growth was furthermore examined. It was found that polyphenol-glycoside, as one of the transfer products, exhibited high antibacterial activity on the growth of Bacillus subtilis and Escherichia coli, no effect when on Bacillus cereus.Key words : Cyclodextrin glucano transferase (CGTase), enzymatic transglycosylation, polyphenol glycoside, antibacterial activity.

Abstract

ABSTRACTMetabolism of Benzonitriles by Flavobacterium sp. NUB 1. Flavobacterium sp. NUB 1 was isolated from industrial waste of PT. Petrokimia Gresik. The bacterium was able to utilize benzonitrile and acetonitrile and propionitril as the sole source of carbon and nitrogen. Growth on benzonitrile gave higher growth rate and biomass yield than growth on acetonitrile and propionitrile. When Flavovobacterium sp. NUB1 grew on benzonitril 15 mM , the doubling time is 9 hours 54 minutes and the specific growth rate (?) was 0,07 h-1. Whole cell of Flavobacterium sp. NUB 1 could hydrolyzed aromatic and aliphatic nitriles. The bacteria isolate has ability in metabolism of acetonitrile greater than benzonitrile. Activity of nitrile hydratase and amidase are more dominant than nitrilase in metabolism of benzonitrile.Key words: Biodegradation, benzonitril, Flavobacterium sp. NUB 1, nitrile-hydratase,amidase, nitrilase
